Bio Statement
My laboratory has a primary research focus on the use of molecular biological techniques to investigate the pathogenesis of invasive ovarian cancer and borderline epithelial ovarian tumors. The laboratory has pursued studies of genetic changes in tissues from invasive ovarian cancer and borderline tumors to better understand the potential relationship between these two entities and to termine basic mechanisms of development. Studies from the laboratory indicate that the pathway of pathogenesis of borderline ovarian tumors is distinct from that of invasive ovarian cancers. Furthermore, we also showed that advanced ovarian cancer is unifocal in origin, thereby indicating the opportunity for early diagnosis. My laboratory’s ongoing research projects involve the use of oligonucleotide microarrays and SNP arrays to identify differentially expressed genes and DNA copy number abnormalities in microdissected primary and recurrent ovarian tumor tissue samples of different histological types, stages, and grades, correlation between expression profile data and clinical outcomes including chemoresponse and survival, and functional studies of genes that may be of clinicopathological significance. These studies will generate biomarkers for the development of a pathway model for the genetic progression of ovarian cancer, provide insights into their roles in ovarian carcinogenesis, provide markers for early diagnosis of ovarian cancer, and new therapeutic targets for treatment.
Besides using oligonucleotide microarrays to identify markers for ovarian cancer, my Laboratory is also focusing on using oligonucleotide microarrays, SELDI ProteinChips, MALDI-TOF/TOF mass spectrometry, and 2D differential gel electrophoresis to identify markers, including RNA, peptides, and autoantibodies, in serum, or plasma samples obtained from ovarian cancer patients. We are developing a new nanochip platform, which can be used to quantify serum/plasma biomarkers.
My Laboratory is also interested in understanding how various endogenous factors such as reproductive and genetic factors, and environmental factors interact to modify ovarian cancer risk. Ongoing research projects involve studies on the effect of various endogenous factors, such as genetic polymorphism, and environmental factors on ovarian cancer risk. In addition, we have recently developed a three-dimensional normal HOSE spheroid Matrigel model, which will be used to study epithelial and stromal cell interaction in ovarian cancer pathogenesis.
In addition, we are interested in the application of targeted nanoparticles to increase efficacy and decrease toxicity in ovarian cancer treatment.
